Flow (Intracellular): 0.25-0.5 ug/10^6 cells Western blot analysis: 5 ug/ml Storage Store at 4°C, stable for 6 months. For long term storage, aliquot and store at -20°C. Recommended Positive Control : Human thymus, RORg transfected cell lysate (IMGENEX, 40211)
Background ROR-gamma is a 560 amino acid novel DNA-binding transcription factor belonging to the ROR/RZR orphan nuclear receptor subfamily. The specific functions of this protein are not known. However, reports suggest that this protein is a ligand activated transcription factor and may be essential for lymphoid organogenesis (lymph nodes and Peyer's patches) and may play an important regulatory role in controlling apoptosis during thymopoiesis. ROR-gamma inhibits the expression of Fas ligand and IL2 and directs the differentiation program of proinflammatory IL-17+ T helper cells. ROR-gamma shares 50 to 51% sequence identity with RORA and RORB and is 88% identical to mouse ROR-gamma. It is highly expressed in skeletal muscle and at lower levels in various other tissues.
Antigen
A portion of amino acids 1-50 of human RORg was used as the immunogen.
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human (isoforms CRA_a) and chimpanzee, 96% homologous in dog, 92% homologous in mouse and rat and 88% homologous in cow. This antibody will detect RORg and may detect RORgt, but can not be used to distinguish between the two proteins.
Western blot analysis of RORg in A) human thymus lysate, B) transfected 293 cell lysate (IMGENEX, 40211), and C) untransfected 293 cell lysate, using IMG-6275A at 5 ug/ml.
Intracellular flow cytometric analysis of RORg in human monocytes using 0.25 ug of IMG-6275A. Shaded histogram represents cells without antibody; green represents isotype control (IMGENEX, 20109); red represents anti-RORg antibody. IMGENEX's intracellular flow kit (10083K) was used for this test, and an anti-mouse IgG1 PE conjugated secondary.
