Background
Principal Names: Xeroderma pigmentosum, complementation group A Official Gene Symbol: XPA Gene ID: 7507 (Human) Gene Map Locus: 9q22.3 (Human). The human XPA (xeroderma pigmentosum group A) gene encodes a protein of 273 amino acids with a zinc finger motif. The protein is an indispensable participant of human nucleotide excision repair (NER) that recognizes and removes a large variety of bulky DNA lesions, which are structurally and chemically distinct. The gene is required for both global genome repair (GGR) and transcription-coupled repair (TCR). In the mechanism of human NER, it interacts with the replication protein A which also recognizes bulky DNA damage, to form a tight protein-protein complex. The protein forms a complex with HR23B, which is generally involved in the initial DNA damage recognition in GGR, probably followed by the further recognition of the damage by XPA-RPA. The protein has also been shown to bind to ERCC1 and TFIIH. The pathological role of XPA has been observed in xeroderma pigmentosum complementation group A (XP-A).

BALB/C mice were injected with recombinant human XPA protein.

Application Notes
We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the end user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Specificity: This antibody reacts with a 40 kD protein known as XPA (Xeroderma pigmentosum group A) protein. XPA specifically recognizes the UV or chemically damaged DNA lesions and triggers the nucleotide excision repair process. XPA binds to the replication protein A (RPA) or excision repair cross complementing 1 protein (ERCC1). Cellular Localization: Nuclear.
