ProVectin™ Protein Delivery Reagent
The ProVectin protein delivery reagent is a unique lipid-based formulation that allows the delivery of proteins, peptides or other bioactive molecules for a broad range of cell types. ProVectin offers the following advantages:

Antibody Plus Provectin
Five micrograms of Provectin was dried out of chloroform into an eppendorf tube. Two micrograms of a fluorescein labeled antibody in 25 microliters of serum HEPES buffered saline was added to the dried Provectin formulation and the solution was vortexed. The mixture was added to the cells, the incubation was continued for four hours at 37°C under 5% CO2, and the cells were examined under the fluorescence microscope to visualize uptake of fluorescein labeled antibody. ProVectin Details

ProVectin is hydrated with the target protein in physiological buffer. During this lipid hydration step liposomes form, and some of the protein that is dissolved in the hydration medium becomes encapsulated in the liposomes; depending on the physical properties of the protein, some of the protein is captured by association with the liposomal membane. The ProVectin/protein liposomes are placed on cultured cells where they attach to negatively charged cell surfaces. Following cell surface attachment the liposomes fuse directly with the plasma membrane and deliver the captured protein into the cell, or alternatively, the liposomes are endocytosed and then fuse with the endosome, releasing the liposome captured protein into the cytoplasm. ProVectin™ is remarkably effective for delivering a number of different macromolecules into the cytoplasm of cells; it can deliver fluorescently labeled antibodies, high and low molecular weight dextrans, phycoerythrin-BSA (300,000 Mol.Wt.), caspase 3, caspase 8, granzyme B, and b-galactosidase into the cytoplasm of a variety of different adherent and suspension cells. The efficiency varies from 20-100% of the cells transduced and up to 50% of the input protein gets incorporated into the cells.


Furthermore caspases delivered to cells with ProVectin TM are functional and drive cells into apoptosis. The robust nature of these results shows that ProVectin is effective for delivering a variety of different macromolecules into many different cell types.

Stability & Storage
The ProVectin reagent is shipped on ice. Upon receipt and for long-term use, store all reagent tubes at –20°C. ProVectin is stable at least for 1 year at the recommended storage temperature.
